I'm using infinix hot 4 pro which was rooted using TWRP/ MAGISK v17.1. Please how do I perform OTA update.
(NB: I don't know how to unroot to perform the update.)
Please kindly help.

You should unroot first.
Unroot by flashing the firmware for your current build number (excluding userdata and cache)

That means I should also flash preloader.
Secondly, will the flashing remove twrp from my phone?

since you're trying to perform an ota upgrade, you could simply flash only stock boot, system and recovery
and yes, flashing stock recovery.img will remove twrp
